DE-96008 DDE/DDT ELISA Test Kit
美国GSI公司的DDE/DDT ELISA Test Kit是一种高灵敏度的竞争型ELISA试剂盒，用于测定样品中的DDE/DDT.ELISA的基础是抗原或抗体的固相化及抗原或抗体的酶标记。

DDE/DDT试剂盒是利用间接竞争酶联免疫检测原理。被检样品和DDE/DDT特异性抗体同时加入到微孔，此时样品中的DDE/DDT和包被在微孔底部的DDE类似物竞争和特异性抗体结合，然后孵育60分钟，孵育结束后倒掉微孔中溶液，清洗微孔除去微孔中没有结合的DDE/DDT与特异性抗体，再加入HRP标记的二抗孵育30分钟，第二次清洗微孔除去没有结合的酶标二抗，加入底物显色液作用20分后用酸终止反应，读取OD值。颜色的深度和样品中DDE/DDT的浓度成反比。

使用步骤：

Step 1. Pipet 100 ul of each pre-diluted positive serum, pre-diluted negative, control (containing only sample diluent), samples containing Toxoplasmosis antibodies (diluted as required) in duplicated wells. Mix gently and incubate at 37 ℃ for 30 min.

Step 2. Aspirate and wash the plate three times. Add 100ul of enzyme conjugate to all wells, mix gently and incubate at 37 ℃for 30 min.

Step 3. Aspirate and wash the plate three times. Add 50 ul of each substrate A and B solution to all wells, mix gently, and incubate in the dark at room temperature for 10 min.

Step 4. Pipet 50 ul of stop solution into each well and mix gently (blue color turns yellow). Measure OD at A630 nm. The samples are positive if the value is > than 0.4, skeptical if the value is between 0.2 and 0.4 and negative if < than 0.2.

安全注意事项

在准备准备样品和对照样品时所使用的人血清，经测试过没有发现表面活性抗原，也没有发现抗体的，而且毒测试呈阴性。但无论*，没有任何测试方法可以保证*不含任何病毒、病毒，毒或者其他病毒，所以这些试剂应被当做潜在的生物危险品处理，并和其他的血样样品一样处理。
