The TMV ELISA kit is based on the sandwich ELISA principle. The kit measures an unknown amount of TMV present in the sample. We used purified Human TMV antibody to coat on microtiter plate wells, resulting in formation of solid-phase antibody. TMV is then added to the wells. An antibody-antigen-enzyme-antibody complex is made by adding a combined antibody labelled with HRP. TMB (3,3',5,5'-Tetramethylbenzidine) is then added as a visualizing agent which gives blue color. Finally, the enzyme-catalyzed reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of TMV in the samples is then determined by comparing the O.D. of the samples to the standard curve.
