背景描述:

Foot-and-mouth disease (FMD) is caused by the foot-and-mouth disease virus which is a single stranded RNA virus from the Picomaviridae family and Aphthovirus genus. It is a very small nonenveloped icosahedral virus. FMD is a very contagious disease and can be death threatened. It can infect cattle, water buffalo, sheep, goats and pigs, but not human. The symptoms of this disease include fever and blister-like lesions followed by erosions on the tongue and lips, in the mouth and between the hooves. In some cases the disease can lead to myocarditis and death especially in young animals. Most animals can recover but they will be harmed and it will influence on the production of milk and meat. After recovery the animal will show no symptoms but it will still carry FMD virus and can transmit to other animals. The incubation time is between 1 to 8 days, but usually it shows symptoms within 3 days. The virus lives in lymph nodes and

bone marrow where the pH is neutral. Depending on the pH and temperature environment the virus can survive for up to 1 month. FMD viruses can be spread by animals, people or materials that bring the virus into physical contact with the animals. 

Genemed Synthesis’s Swine Foot and Mouth Disease Distinguishing ELISA Test kit is a highly sensitive indirect type assay for the detection of FMD-3ABC antibodies in the serum, differentiation naturally infected from vaccinated swine.

使用步骤：

Step 1. Pipet 100 ul of each pre-diluted positive serum, pre-diluted negative, control (containing only sample diluent), samples containing O type antibodies of Porcine FMD(diluted as required) in duplicated wells. Mix gently and incubate at 37 °C for 30min.

Step 2. Aspirate and wash the plate three times. Add 100ul of enzyme conjugate to all wells, mix gently and incubate at 37 °C for 30min.

Step 3. Aspirate and wash the plate three times. Add 50 ul of each substrate A and B solution to all wells, mix gently, and incubate in the dark at room temperature for 10 min.

Step 4. Pipet 50 ul of stop solution into each well and mix gently (blue color turns yellow). Measure OD at A630 nm. The samples are positive if the value is > than 0.4, between 0.2 and 0.4 is skeptical and negative if < than 0.2.
